Issue:
When trying to run a method, the Agilent Fragment Analyzer controller software shows the message Setup incorrect - Ladder file does not exist (see Figure 1).

Resolution:
The software displays the error message because the check box Use size calibration file is selected (marked in Figure 1), which requires the user to load a .SCAL ladder file. If the Size calibration file path section is left blank, this error message is displayed.
If it is the first time you are running this method, you will need to run a ladder in the sample plate. For this initial run, select Create size calibration file in the Separation Setup dialog box and a .SCAL file will be created when the run completes. This .SCAL file can be used for future runs with the same method. Alternatively, you may export a ladder file from an existing run through ProSize (open the run and click Analysis > Show size calibration > Export).
For future runs, you wouldn’t need to run a ladder and can select the Use size calibration file check box. Then load the saved .SCAL file by clicking the three dots behind the Size calibration file path section (see Figure 1), which will be used for analysis of the run in ProSize.
Caution: Use the saved ladder for no longer than one week. The runs you are importing the saved ladder into should be run with reagents from the same vials.
