Issue
Is it possible to analyze single stranded DNA (ssDNA) on the TapeStation? Which kit is the most suitable for analysis of ssDNA?
Resolution
Agilent does not offer any kit dedicated or validated for ssDNA analysis. None of the kits have been tested for performance with ssDNA.
However, given that the TapeStation RNA assays are validated for total RNA, which is a single stranded nucleic acid, the RNA kits can be considered. The dye used in the RNA kits has a higher affinity for single stranded material than the dye used in the DNA kits.
The migration profile, sizing, and quantification might not be accurate for ssDNA because the RNA kits were validated for total RNA samples. Agilent cannot guarantee that data obtained for ssDNA using RNA kits is within the given specifications of the respective assay.
Caution: Agilent does not support and has not validated the examination of ssDNA samples on the TapeStation.
