Can I run qPCR samples containing Sybr® Green I on the Bioanalyzer?
Background
Sybr® Green I is an Intercalating Dye commonly used to detect amplification products in real-time PCR. This dye interacts with single and double stranded DNA in a nonspecific manner. It shows an increased fluorescence intensity upon binding to DNA.
Resolution
It is possible to run and detect qPCR samples on the Bioanalyzer even in the presence of Sybr® Green I. Amplicons containing Sybr® Green I can be detected without disturbances using the DNA 1000 assay on the Bioanalyzer.
Caution: The incorporation of Sybr® Green I into the sample results in a small but significant increase of fluorescence intensity and a small but significant increase in the electrophoretic mobility. These changes must be considered when running samples containing Sybr® Green I on the Bioanalyzer.
