  • What to do if my LC-QQQ Autotune Fails?

This Information Applies To: Agilent 6400 Series triple quadrupole LC/MS systems Issue Autotune fails to complete. Background Agilent 6400 LC/MS systems use an Autotune algorithm to optimize the sensitivity and mass resolution of the instrument. Sometimes when running the autotune procedure the autotune does not complete and the user is given an Autotune failed message in MassHunter. One of the main reasons for Autotune failure is a lack of sensitivity or abundance of the tuning ions...

How to Reset the Agilent GC/MSD Tune File to Default Values

Answer

This Information Applies To: Agilent GC/MS MassHunter Acquisition Software

Issue

If there is a problem with the current tuning parameters and a need to tune from the default parameters, see the following procedure to reset the EI tune file (atune.u) to the default values.

Steps to follow
Method 1
  1. Locate the current tune file in Windows File Explorer under the path x:\Masshunter\GCMS\n\5977 (x is the installation drive of MassHunter folder, n is the instrument number). Rename the atune.u tune file ( e.g. atune_old.u ).

  2. Locate and Copy the default tune file (atune.u) using Windows File Explorer from path C:\GCMS\MSSETUP\5977, and paste it into the same location as the renamed tune file in Step 1.

  3. The copied default tune file needs the MS polarity set in Manual Tune view (View>Tune and Vacuum Control > Parameters > Manual Tune). Save the tune file with the correct polarity for your instrument. The polarity information will be in the previous tune reports but also confirm it on the instrument.

In Figure 1, see the “X” symbol on the right side of the “Neg” box on the MS analyzer label – it means that negative polarity is to be set for this instrument.

Figure 1. MS Analyzer label – Negative Polarity

  4. Tune the MS before data acquisition after resetting the EI tune file (atune.u) to the default values.

Method 2
Updating or refreshing the instrument configuration in the GC/MS Configuration editor will reset the EI tune file (atune.u) to the default values.
  1. Close the GC/MS Acquisition software and open the Agilent GC/MS Configuration editor.
  2. Click the instrument number on the left at the top to open the instrument configuration.
  3. Do not change any information, just click Ok, when the dialog box opens "proceed with update to instrument configuration," click Yes.
  4. The update is now done, close the Agilent GC/MS Configuration editor.
  5. Tune the MS before data acquisition after resetting the EI tune file (atune.u) to the default values.
The following video (Figure 2.) demonstrates how to update GC/MSD configuration as described in Method 2.
