Can I Mix and Match TapeStation Reagents and ScreenTapes?

Answer

Issue

Can I run an assay using the ladder or sample buffer from a different assay if I have run out of the appropriate assay ladder or sample buffer? Is it possible to run an assay using a ScreenTape device of one assay type with the reagents from a different assay? 

Resolution

It is not possible to use the sample buffer or ladder from a different assay type for the following reasons:

  • The salt concentration in the sample buffer is assay-specific. If the wrong sample buffer is used, the migration of samples through the gel will differ due to the differing salt concentration.
  • The assay markers are included within the sample buffer. The concentration and size of the markers is assay-specific and cannot be altered in the software during the analysis. The markers are used to align the samples and are therefore important for correct sizing and quantification. If the incorrect sample buffer is used, the size and concentration of the markers will differ from the size and concentration in the analysis software, and therefore the sample concentration will be calculated incorrectly. 
  • The number of ladder peaks and the ladder peak size and concentration is assay-specific. The ladder is used to calculate the sample size. Sizing will be incorrect or unavailable if the incorrect ladder is used.

It is not possible to use a ScreenTape device of a different assay type for the following reasons:

  • The ScreenTapes are barcoded and determine the assay that the instrument will run. The electric current applied during each assay is unique. If you use the wrong type of ScreenTape, the electric current applied to the sample will be incorrect.
  • The gel in the ScreenTape is not the same in every assay. If you separate your sample with the wrong ScreenTape, it may not separate as expected through the gel.
  • Once the ScreenTape barcode is read, the software expects certain parameters such as the buffer to sample ratio.
  • The ScreenTape determines what analysis settings are applied to the data file after the run has finished. If you use the wrong ScreenTape device, the software may expect a different number of ladder peaks. The anticipated concentration and/or size of the ladder and marker peaks, and therefore the sample concentration and sizing, would be incorrect.
