Creating an AriaMx Custom Template

Issue

Creating an AriaMx custom template that is accessible from the My Templates section of the AriaMx software.

Background

When running the same assay or similar assays repeatedly, custom run templates can significantly streamline your workflow. These templates allow you to start with predefined plate setups and thermal profiles, saving time and ensuring consistency.

Steps to Follow

1. Create a template:

    • Open the AriaMx software.
    • On the getting started page select File > New (Figure 1).
    • The Experiment Types page will appear. This page has a box where you can enter a name for your new template file (Figure 2). Choose a name that is relevant and easy to remember to help you identify the template in the future. 
    • Click Create.

2. Complete the Plate Setup:

    • Set up your plates according to the requirements of your assay ensuring that all necessary wells, controls, and samples are correctly positioned.

3. Define the Thermal Profile:

    • Configure the thermal cycling parameters (e.g., denaturation, annealing, extension) for your experiment.
    • Specify the temperature and duration for each step.

4. Save as a Template:

    • Once your plate setup and thermal profile are ready, click File > Save as Template.
    • When prompted, save it in the following location: C:\Users\Public\Documents\Agilent Aria\Experiment Templates.
    • Confirm the name of your template, e.g. Custom qPCR template.amxt.
Your new template will now be listed in the My Templates page within the software.

Visual Overview

For a clear overview of your custom templates, follow these steps:

  1. Open the AriaMx software.
  2. On the getting started page select File > New (Figure 1).
  3. Select My Templates from the left side of the software screen.
  4. Click the Tile View icon in the top-right corner (Figure 3).
  5. This view will display a graphic representation of the plate layout and the thermal profile for each template.
