This Information Applies To: Agilent AriaMx real-time PCR system
Issue
When configuring a plate setup in the AriaMx software, it is crucial to ensure that the selected dyes correspond to the optical modules installed in the instrument.
Background
If there is a mismatch, the following error message will appear in the AriaMx software while priming of the experiment: The dyes configured in plate setup do not match with the optical modules detected at the instrument. Reconfigure the plate dyes at the instrument and try again (Figure 1).

Resolution
To identify the discrepancy, return to the plate setup screen in the AriaMx software. The filters not present in the AriaMx will be highlighted in red. The dyes that are selected for use in the assay will have a tick in the check box to the left of each dye name (Figure 2). If a dye has a tick in its check box and is highlighted in red, the dye is selected in the experiment, but the corresponding optical module is not installed.

To resolve this issue, you have two options:
After you have installed the required optical modules or changed which dyes are selected, prime the experiment.
