Using the Method Translator Calculator in the HPLC Advisor App

Issue

Legacy HPLC method needs to be translated to a new column or LC system (HPLC to UHPLC or Analytical to Preparative LC).


 

Requirements

Install the most recent version of the HPLC Advisor app, see Installing the HPLC Advisor App.

Steps to Follow

Open the method translation calculator

  1. In the app home page, press Calculators.
  2. Press Isocratic Method Translation or Gradient Method Translation according to your method (See Figure 1). The gradient method translation is used as example in this article.

Original method

  1. Type in the parameters of the original method, including original column geometry, LC system dwell volume, flow rate, and injection volume (see Figure 2).

  2. Scroll down to Your original gradient profile section, and edit the current steps by pressing the pencil icon on the right, or press the + Add step button. 

  3. Type the Time and %B for the gradient step. Press Save and add another step +, to continue adding gradient steps, or press Save and close to finish (See Figure 3).

  4. Press Next.

 Tip 
Tip: To get more information of a parameter, press the information Information source icon on the top right of the parameter field

 

Translated method

  1. Type in the parameters of your translated column geometry, and LC system dwell volume.

  2. The flow rate, injection volume and gradient profile of translated method are calculated and shown immediately.

  3. The performance of translated method is also shown comparing to the original method at the bottom (see Figure 4).

  4. The calculation is finished, and the data can be saved or exported to PDF. If you have a preferred flow rate for the translated method, press Modify.

Modified translation profile (optional)

This step is optional, only if you need to use a certain flow rate for the translated method. 

  1. In the Translated method section, scroll down the bottom of the screen and press Modify.
  2. Set the preferred flow rate.
  3. Modified gradient and performance comparison are calculated and shown (see Figure 5).

Save data or export as PDF

Press the vertical ellipsis icon on top right of the calculator screen to save data or export as PDF (see Figure 6).
