asking for your opinion on optimizing

Dear. LC/MS scientists 
 I truly need your help. I have optimized ginsenoside Compound K (C36H62O8, 622.44) for 2 weeks. I was using MH Optimizer program. I confirmed that the precursor ion and product ion was same as article (621.4/161). The injection condition was 10 ug/ml, 2 ul. I use the negative method. precursor Ion abundance was 60000, product Ion was 1500. I know that negative conditions are generally lower than positive conditions. But I can&rsquo;t figure out why I can&rsquo;t check the linearity. How can I solve this problem? 
 I'm sure it's not a hardware issue because the machine was inspected two weeks ago.

Hi Chanil.Park , 
 I think your ce value may be too high. Did you try manuel optimize, your compund. Does the area decrease as the value of ce increases? After that in MRM mode , you can verify the ce value.
