I run the sequence but when I opened the GC Offline to check the nicotine level it didn't pick anything. It wasn't even showing zero just blank. How can I resolve this issue. I am attaching picture as reference.

From the picture, it appears your FID flame was extinguished. You can put a metal wrench or a mirror at the top of the detector to see if the exhaust contains moisture from the combustion, but a flat baseline at zero output usually means the flame is out.
