This Information Applies To: 2100 Bioanalyzer, Bioanalyzer chips Issue: Can 2100 Bioanalyzer assay chips and reagents be interchanged? Resolution: Some assay chips can be used with multiple assays, however some cannot because the chip layout and assay script are different. Caution : Agilent does not guarantee the proper performance of runs when chips and reagents are interchanged inappropriately. Table 1 shows the range of Agilent Bioanalyzer assay chips available...

This Information Applies To: 2100 Bioanalyzer system, 2100 Expert software Issue: How to identify and troubleshoot late migration of the ladder, marker, and sample peaks in data generated on the Agilent 2100 Bioanalyzer system. Background The ladder, marker, and sample peaks in the affected run take considerably longer to migrate compared to the peaks in the demo file of the same assay type. Late migration is usually accompanied by errors, similar to the ones shown in Figure 1 . These...

This Information Applies To: 2100 Bioanalyzer, 2100 Expert software Issue The Agilent 2100 Bioanalyzer system electropherograms in the Agilent 2100 Expert software have a wavy, uneven baseline ( Figure 1 ). Figure 1. Wavy, uneven baseline on Bioanalyzer electropherograms The error is most often associated with: Problems during chip priming Dirty electrodes Moisture on the electrodes Sample contamination The sample concentration being higher than the assay's range Liquid spillage...

This Information Applies To: 2100 Bioanalyzer, 2100 Expert software Issue The Agilent 2100 Bioanalyzer system run ends prematurely and the following error appears in the logbook: Run aborted on port x. Instrument error occurred on port x, Unusual high or low voltage or current was detected during the start phase of the on-chip analysis. The marked wells will indicate the wells on the chip that caused the problem. An example of the error is shown in Figure 1 . The error is most often associated...
  • Agilent 2100 Bioanalyzer System Chip not Detected

This Information Applies To: Agilent 2100 Bioanalyzer system, 2100 Expert software Issue: The Agilent 2100 Bioanalyzer system does not detect a prepared microfluidic chip after it is inserted into the instrument. The interface shows the instrument icon (or the last chip type used). However, the start run checklist shows that a chip is not detected (see Figure 1 ). Figure 1 . Chip not detected (see red arrow) Not detecting a chip can indicate an issue with the Bioanalyzer-PC connection...

How to Run the Hardware Diagnostics on the Agilent Bioanalyzer

Answer

This Information Applies To: 2100 Bioanalyzer, 2100 Expert software

Issue:

Running the hardware diagnostics on the Agilent 2100 Bioanalyzer system can help to investigate whether the instrument performs as expected.

Steps to follow:

Required material

  1. An empty clean unused chip of any type (DNA, RNA, or protein)
  2. The test chip kit (p/n G2938-68300) consisting of an electrode / diode test chip and an autofocus test chip
Tip  Tip: Verify that the test chip kit is within the expiry date and the test chips have been stored in their original bag. Avoid touching the underside of the test chips.

The Bioanalyzer Troubleshooting and Maintenance Guide provides an overview of the diagnostics in the Hardware Diagnostics section.

  1. Start the Agilent 2100 Expert software.
  2. Access the hardware diagnostic tests by selecting the Diagnostics tab in the Instrument context of the 2100 Expert software (Figure 1).
  3. In case multiple 2100 Bioanalyzer instruments are connected to the PC, highlight the appropriate instrument in the tree view.
  4. Select any of the hardware tests from the list or choose Select All to run all tests.
  5. Select Start and follow the instructions as given by the 2100 Expert software.
  6. If there are any failures, repeat the failed tests in isolation.
  7. If the failures persist, send the diagnostic test results file to Agilent technical support in the country where the instrument is located. The results of diagnostic tests are stored as .xdy files in C:\Program Files (x86)\Agilent\2100 bioanalyzer\2100 expert\diagnosis. Alternatively, create a Support Package by clicking Help > Create Support Package.... The File Collect dialog box will appear. Select all check boxes to avoid useful information being missed. Clicking Collect will generate a support package and save it to your desktop (Figure 2). This support package will contain the diagnostic results and should be sent to Agilent technical support via email.

Resolving common test failures

Short circuit test

  • If the failure occurs after electrode pin set cleaning, make sure that the pin set is completely dried before rerunning the test. Remove the pin set from the electrode cartridge and dry both with oil-free compressed air thoroughly. Alternatively, let the pin set dry overnight.
  • If the pin set wasn't cleaned before the diagnostics, perform the maintenance of the electrode cartridge as described in the Bioanalyzer Troubleshooting and Maintenance Guide on page 155.
  • Ensure that the humidity level in the environment is below 50% and the ambient temperature is 25 °C.

Optics test

Clean the focusing lens with lens-grade, lint-free tissue paper, and isopropanol before rerunning the test.
