Hi Ricky,

We do not recommend using this media for culture, only for assays. This is because the assay media will be missing many components that are necessary for cell culture (sodium bicarb, substrates, etc.).

I would like to use it adding SCF,IL-3, EPO, lipids, glutamine, pen/strep and transferin as typical growth factors for CD34+ cells. I would add different concentrations of glucose. I have noticed there is HEPES and even different amminoacids included in the formulation but they're not enough I guess. HEPES amount especially is too low. Tipically it should be around 25 mM.

Unfortunately, we would still not recommend doing so as this media has not been validated for the longer conditions of cell culture, only shorter Seahorse assays. I am sorry about that!
