Recently, I am trying to applying the method 5990-7950EN to the diet sample and raw material sample. But when I finished my sample preparation and run my sample on the HPLC. I found the peak for the vitamin b5 is higher and wider than I expect. I am wondering if this method is to measure the total pantothenic acid or the free pantothenic acid. One explanation for that is the peak area for B5 is the sum of the CoA, panthenie and free pantothenic acid. This may be cause pantothenic acid has different forms in the food. But in vitamin tablet sample the peak is quite clean. So I hope to get some suggestions! The attached photo is the comparison between the food samples and tablet samples
