I am running a hydrolyzed gluten sample and I want to determine each gluten fragments after hydrolysis. I developed a method shown in the file attached here and tried to run based on this method one representative alpha chymotrypsin digest gluten. The result I got is less than I expected as compared to other literature. I thought maybe I missed some important parameters in the method development and I want your recommendation to come up with a good result.
