Sorry for the delay. The compound math column was not introduced until rev B.07.01. Probably the best thing for you to do would be to upgrade to a more recent revision of the software so that you can take advantage of the workflow outlined in this video: How to summate peaks in MassHunter Quant B.08.00
If you are unable to upgrade, then this document walks through an alternative, more involved workflow: Instructions for Enabling Compound Math on B.03.0x-B.07.00

With respect to the instructions to enable the compound math, I have the following questions:
Item 4) Save the file
If I have compound 1, compound 2; and compound 1 + compound 2. Why is compound 1 / compound 2 added?
In Custom Calc I obtained a zero value and the responses of both compounds were not added.
Something I'm doing wrong?

Hello,
Sorry for the delay.
I initially thought that maybe the spaces in your compound names were causing an issue, but I was not able to reproduce this issue. Is it possible that you did not copy another compound's calibration levels to the new "Math Compounds" when you added them to the method?
