I run on an ALS, so manual injections aren't really my bag. But , if you look at your sequence you should see a column labeled something like "injection source". Is the entry in that column "manual injection"? You have, I believe, three different choices: one "per method", one "gc injection" and one "manual injection". That might be your problem.
I usually run on ALS but don't have one for the current instrument in question. I looked at the injection source and only see external or no injection/instrument blank.
Go into your sequence table and check out the injection source column and see what options it gives you. Mine give me this:

Sounds like you're using OpenLab CDS not Chemstation. I would have expected selecting External would allow you to do manual injections. I can't work out what sort of instrument your using.

by saying ChemStation to OpenLAB CDS I meant that I was transitioning from one to the other. I. was able to select external injection on the sequence and initiate it by pressing start on the instrument keypad. thank you.
