Scan Mode

Question

Hi guys, I am using an LC-QqQ-MS/MS 6460c equipment and I'm having issues with the scan mode, both in positive and negative mode. When I inject a solution in a method using MRM, it analyzes correctly. However, when I try to characterize the compounds either in SCAN mode or even in the optimizer, the equipment does not recognize and does not work correctly. What could be the issue?

 For scan or SIM, which is typically what the optimizer starts with, you will typically need to use a higher concentration of your sample in order to see signal. You could try extracting EICs from your scan data to see if you are able to find any target signal, but if not, I would start slowly increasing your concentration by 5 to 10x until you are able to see your target in full scan mode. Once you can see it in scan, then it should be detectable in optimizer/SIM mode.
