Combining the SRM responses for two internal standard compounds to produce a single 'internal standard' used for quantitation purposes with MassHunter Quant software

Is it possible with MassHunter quantitation software to combine the SRM peak responses for two internal standard compounds, for example with PFAS analysis, 13C2-PFDoDA and 13C2-PFTDA, and use the resulting combined response as an internal standard called ‘13C2-PFDoDA (C12) + 13C2-PFTDA (C14)’ for PFTrDA (C13) quantitation?

If it is possible, how is it done?

  • In Method Set Up Tasks > MRM Compound Setup, Add the two columns, 'Cmpd. Group' and 'Compound Math' if they're not already present. In the 'Cmpd Group' box for the two ISTDs that you want to average add for example "1" in each box. Then add a new row for an ISTD that for example combines the names of the two ISTDs that you want to average. Add the same name in the 'Cmpd Group' box that you used for the two ISTDs that you're averaging, in this example "1" and in the 'Compound Math' add 'Response Average' from the dropdown menu.
