System Tune LC/QTOF 6545XT

Hello everyone,

I am facing an issue when trying to perform a System Tune in negative mode of both Quadrupole and TOF following 1 month of the QTOF being off due to a hard drive failure.

Has anyone encountered that error and was able to fix it ?

  • Hi,

As per the autotune record, the detector MCP is maxing out at 1000v. It cannot get enough signal to achieve the detector calibration.  You can see that from the beginning at line #19 where detector MCP jump from previous 720v to 900v.

Line #19  Signal of the Highest m/z Peaks Below Requirement, Increase MCP to 900

Line #60 Searching MCP to Produce 20 Counts per Single Ion

               Counts         MCP     Frequency               Status

               08.5              936        8.1 %                in Progress

               09.8              971        7.8 %                in Progress

Line #64  Trying to set MCP to 1001.79694352159, which is out of bounds, set to maximum: 1000

               11.6              1000        9.4 %                in Progress

Check the basic first, enough tune mix solution in bottle, inspect nebulizer needle for physical damage and proper adjustment.  Check the tune mix flow out of the peek tubing on top of nebulizer, disconnect the tubing, with bottle B to On, you should have 1 drop falling off every 7-10sec.

Check the result in positive mode from MCP value.  If it’s close to max 900-1000v, like negative mode that may indicated the detector end of life, may need replacement.  But If your Positive mode passes with normal MCP value750-850V, but the Negative mode failed, that may indicate issue with negative mode ion generation at the front end of the QTOF.  I suggest you place a service call at Agilent in both cases.

    Best regards

  • Thanks for your help.

There seems indeed to be an issue linked to the detector: after testing it in Positive mode, the tuning doesn't pass and MCP is maxing out as well.
In any case, I will contact Agilent.

    Best regards
