My lab recently installed the PFAS conversion kit on our 1290 Infinity II UPLC.
Since we have bypassed the degasser, we are now having issues with (what appears to be) air building up within the system during the course of a run. We degas the solvents via sonication before use. 25% MeOH has also been added to the aqueous phase, and the composition of the gradient has been adjusted accordingly. Pressure fluctuations start to present themselves and progressively get larger throughout the course of the run. Eventually, a critical mass of sorts is reached, and the system is no longer capable of maintaining a consistent pressure at all.
How can I prevent this from happening? Is there a system preparation step that I am missing? I have tried the various "Prepare Pump" functionalities, but they don't seem to be a solution for a longer worklist.
