BioLC SEC - AdvanceBio SEC 200A 1.9um 2.1x150mm PEEK

Hello Community,

I was running the above mentioned HPLC column over a period of 12 hours in PBS, and all of a sudden the column went massively over pressure and no longer runs. The PBS was two weeks old, and had been stored at room temperature. Have you ever heard of this?

The recommended cleaning techniques have not improved the situation, and agilent does not recommend reverse flow (which for a blockage of a FPLC column is something that one would do). Any suggestions would be highly appreciated,

  • Two weeks at ambient temp and possibly not 100% light protected can lead to biological growth in PBS. If that happens the inlet frits are totally blocked (maybe even some frits in the system). If it is the guard column that is blocked, you have to replace it.

    Be aware that 1.9 µm materials have very low porosity frits (ca. 0.5 µm). Unfortunately column frits cannot be replaced and, as you already mentioned, the column cannot be reversed.

    For future reference, you should filter your buffer/eluent at least twice a week and do not use it longer than a week.

    For possible cleaning solutions, check the user manual.
