Low signal and no ladder with RNA Pico 6000 kit

Hello,

I've been having issues obtaining a reliable output from the Pico 6000 kit with RNA I've previously checked via spec measurements; the concentration used is within the acceptable limits stated in the Pico 6000 manual.

However, I'm not getting anything from the ladder. I've prepared it as stated in the instrument manual and ordered a fresh ladder aliquot to retest (results below). All sample material (including the ladder) were kept on ice during chip preparation.

The signal also appears to be very low compared to results I've seen elsewhere using the same kit. One chip out of this lot provided signals of ~2500 and ~250 upon dilution of the same linear RNA sample I'm running here in the last well, but the results cannot be replicated.

    

Would you be able to help me make sense of what the problem might be? I'm happy to provide the data file associated with the run as well if you need it.

  • Sorry to hear about this. Please could send the data file where you experienced this? This would be a file with a .xad extension. By default these are saved in this location: C:\Program Files (x86)\Agilent\2100 bioanalyzer\2100 expert\Data.
