AriaMx Software Converts Standard Amount when Units are Changed

Issue

AriaMx software automatically changes the values of standard solutions.

Background

When setting quantity values for standard wells on the Plate Setup screen, select the Units (for Plate) (e.g., micrograms, nanograms) first, then enter the Amount. If you change the units after entering a value, the AriaMx software will automatically convert the amount to maintain the same concentration.

 For example, if you enter 500 nanograms and then select picograms as the unit, the software will convert it to 500,000 picograms (Figure 1). To ensure that the values remain consistent, select the units for the replicate set of standard wells first, then enter the starting amount.

Steps to Follow

The workflow for assigning replicate numbers and starting amounts to standard wells is described in the following steps and shown in Figure 2.

1. Select the wells that will contain the standards.
2. Select Standard from the Well Type drop-down menu.
3. Select the dye used and give the target a name.
4. Assign the standard curve replicates. Using the Auto Increment button makes assigning replicates is made quicker.
5. Select all the standard curve wells.
6. In the Amount field, type in the quantity of the standard target present in the first replicate set of standard wells.
7. In the A factor of drop-down menu, select the dilution factor used to generate the dilution series.
8. In the Units (for Plate), select the units of concentration for the quantity entered in step 6.
