Quantitation Calculations in OpenLab CDS

Issue

This article outlines different ways to quantify the amount of compound in an unknown sample.

Background

Quantitation uses peak area or height to determine the amount of a compound in a sample. A Quantitative Analysis involves many steps that are briefly summarized as follows:

• Analyze the sample containing an unknown amount of the compound to obtain the response due to the unknown amount.
• Compare the response of the unknown amount to the response of the known amount to determine how much of the compound is present.
• To obtain a valid comparison for the unknown sample response to that of the known sample, the data must be acquired and processed under identical conditions.

Resolution

OpenLab CDS offers the following calculation procedures for determining the amount of each component present in a mixture:

1. With calibration curve:
   1. External standard (see article on How to Create a Multipoint ESTD Calibration Method with Agilent OpenLab CDS (ver 2.x))
   2. Internal standard (see article on ChemStation External and Internal Calibration Calculations)
   3. Indirect quantitation using a calibration compound (see article on Quantitation of Uncalibrated Compounds Using Reference Compounds in OpenLab CDS)
2. Without calibration curve:
   1. Area or Height percent (Area% or Height%) (see article on Area% and Height% Calculation in OpenLab software) 
   2. Quantitation using a manual factor or fixed response factor (see article on Quantitation of Uncalibrated Compounds Using a Fixed Response Factor in OpenLab CDS)
