Setting Up the Mode of Calibration in OpenLab ChemStation and OpenLab CDS

Issue

Finding where to select the mode of calibration with OpenLab ChemStation and OpenLab CDS. 

Background

There are six calibration modes available:

1. Percent: calculated with the relative size of all integrated peaks. There is no calibration curve. The signal can be Area or Height.
2. ESTD: External Standard calibration using Area or Height to generate the curve.
3. ESTD %: External Standard calibration using the percentage of the injected sample, needs a sample amount. The signal can be Area or Height.
4. ISTD: Internal Standard calibration using a ratio between compound and ISTD. The signal can be a ratio of Area or ratio of Height.
5. ISTD %: Internal Standard calibration using a ratio between compound and ISTD reported in a percentage of the injected sample. Needs a sample amount. Signal can be a ratio of Area or ratio of Height.
6. Norm %: calculated has an ESTD curve except there is an extra step to normalize with the other compounds. Only use the normalization report if all components of interest are eluted and integrated. Excluding selected peaks from a normalization report will change the reported results in the sample.

Steps to Follow for OpenLab ChemStation

1. Load the data by double-clicking the sequence name in the list on the left (Figure 1).

2. Click the Report drop-down menu > select Specify Report (Figure 2).

3. Click Quantitation settings (Figure 3).

4. Click the drop-down menu beside Calculate and select the appropriate calibration mode (Figure 4).

Steps to Follow for OpenLab CDS

1. Create a calibration table.
2. Go to Processing Method > select Calibration > select General > select External standard or Internal standard (Figure 5).
   • To do ESTD or ISTD, select either External standard or Internal standard, respectively. 
   • To do Norm%, select the option Normalize to. Users can select %, ppt, ppb, or ppb and give a number. 
   • To do ESTD% or ISTD%, select the option Calculate mass %.

3. Go to Processing Method > select Calibration > select Calibration Table > select Area, Height, Area%, or Height% from the drop-down menu (Figure 6).
